Imagine replacing a missing tooth with a new, natural one

Losing a tooth is frustrating and expensive. Today’s solutions—implants and dentures—work well but do not feel like the real thing, and a single implant can cost US $3,000–5,000. But what if you could regrow your own tooth instead of relying on synthetic replacements?

A team from Kyoto University and the University of Fukui discovered that a protein called USAG‑1 acts as a brake that stops us from growing more teeth after our adult set comes in. By blocking this protein with a monoclonal antibody, they prompted mice and ferrets to grow complete new teeth. In essence, they woke up a third set of teeth hidden in our DNA.

How does the drug work?

USAG‑1 inhibits two key pathways—BMP and Wnt signals—responsible for tooth development. The drug is an antibody that binds to USAG‑1 and blocks its action, allowing BMP and Wnt signals to reactivate and guide the formation of a new tooth. It’s a cell‑free therapy, simpler than complex tissue engineering.

From animals to humans

After success in animals, Kyoto University Hospital launched a Phase 1 clinical trial in 2024. Thirty men aged 30–64 missing a molar received an intravenous injection of the antibody. This stage focuses on safety and dosage. As of April 2026 there were no serious side effects and researchers saw signs of tooth bud activation. If results remain positive, the next phase will test children aged 2–7 with congenital anodontia—a complete absence of teeth.

Scientists believe this tooth regeneration drug could become a third option alongside dentures and implants. Initial costs might be around US $11,000 per tooth, but prices should fall over time.

What about other studies?

USAG‑1 research is not the only path to regenerative dentistry. At Harvard’s Wyss Institute, a team used low‑power lasers to stimulate dental stem cells and form dentin, the hard layer beneath enamel. Instead of blocking a gene, this technique activates a protein called TGF‑β1, prompting the cells to repair the tooth. Though still experimental, it shows how many scientists are exploring ways to help teeth heal naturally.

FAQs

When will I be able to regrow my teeth?

The Japanese trial’s first phase ends in August 2025. If it proves safe and effective, researchers will test it on children and then on adults who have lost teeth due to decay or injury. Experts estimate that the treatment could reach patients around 2030

The therapy involves a single injection. Afterward, the new tooth grows over several months. Animal studies reported no pain, and the resulting teeth were healthy and functional

Initially, the therapy targets congenital tooth loss. Over time, it may apply to teeth lost from cavities, gum disease or trauma. The key is having a dormant tooth bud that can be reactivated.

Like any new therapy, long‑term effects are unknown. Scientists monitor whether regrown teeth align properly, develop roots and integrate with nerves and blood vessels
